The classical complement pathway requires antigen-antibody complexes for activation (specific immune response). C3-convertase cleaves C3 to the active forms C3a and C3b. C3a is a peptide mediator of inflammation (intermediate activity). C3b binds covalently to the microbial surface, acts as an opsonin, and binds to C5-convertase (C3b2Bb). C5-convertase cleaves C5 to C5a (11 kDa) und C5b (190 kDa). C5b initiates the membrane attack pathway, which results in the membrane attack complex (MAC) (= C5b + C6 + C7 + C8 + C9) on the pathogen surface.
