Butyrate kinase is an enzyme that plays a crucial role in butyrate metabolism. It is transcribed from the gene buk and is part of the ASKHA superfamily. The enzyme catalyzes the chemical reaction ADP + butyryl-phosphate ⇌ ATP + butyrate, with ADP and butyryl-phosphate being the two substrates of this enzyme.

The distribution of butyrate kinase is not widespread among human butyrate-producing bacteria. Significant enzymatic activity and amplifiable gene sequences corresponding to butyrate kinase could only be found in a limited number of strains.
