NOD-like receptors (NLRs) come together to form oligomers and connect with adaptor proteins that contain a caspase-recruitment domain (CARD). This domain is a short module responsible for protein-protein interactions, which facilitates the recruitment of Inflammatory Caspases (group I caspases).
In the process of recruitment and activation, caspases remove the C-terminal repressor domain from gasdermins. This action releases the free N terminus, which then creates large pores in the cell membrane.
